Its first minister, the Rev Francis Close, who later became the Dean of Carlisle Cathedral, was instrumental in its establishment. The church was inaugurated with an address from the esteemed Rev Charles Simeon, setting the tone for a legacy of impactful ministry.

Throughout its history, Trinity Cheltenham has faced challenges and triumphs. In 1976, the church was on the brink of closure, but under the guidance of Canon Lawrence Totty, a retired missionary, it began to flourish once more. This revival continued under subsequent vicars, Rev John Risdon and Rev Paul Harris, who expanded the church's reach into the community.

In 1994, a significant transformation began under the leadership of Mark Bailey. The church underwent major renovations, and by 2000, it had acquired Trinity House, a three-story building that would support its growing congregation. The Trinity Growth Project, launched in 2005, further expanded its facilities, including the purchase of the Fusion Building in 2008 and a refurbishment of the main church building in 2009, increasing its capacity to nearly 650.
